Hi, I will be pleased to share any results or experiments that I am working on with people interested. I am a scientist by training and I recently got a lab position at my university. However, since most of my experiments require a lot of time and money, I decided to start some projects to make my time easier and more focused. One of my projects is a FRET-based biosensor for intracellular signaling molecules. The sensor is targeted to the extracellular domain of the receptor. It is equipped with a FRET pair (Cy3/Cy5) to enable a ratiometric FRET biosensor. Also, the sensors contains two fluorescent proteins for validation (EGFP for cell visualization) and two differentially expressed proteins (ERK1 for activation and PTP1B for inhibition). The resulting construct (intracellular sensor) is then delivered into the cells using liposomes (for stable expression).
Now, the output is a live-cell based biosensor using very easy methods. However, once the sensor is expressed, there is a new problem that needs to be addressed before we can obtain quantifiable data. This problem is photobleaching. We cannot image the cells using the FRET pair in presence of light, due to the high energy absorption by the fluorophore at that wavelength. My lab is currently working on developing a special cell line that will not produce new fluorophores for a pre-defined period of time. Also, we are collaborating with another lab that is working on a flow cytometry based method for quantification. Another possible solution is to develop a flow-based method (for sensitive quantification) for intracellular protein concentrations.
I am open to any suggestions, comments, and results, especially if you are using your own sensor (no need to publish results). Also, if you are using a different approach to solve the photobleaching problem, I would be glad to compare the results with yours. Thanks.
